Recent reports from the World Health Organization (WHO) confirm new outbreaks of hantaviruses[1] and Bundibugyo virus (Ebola)[2]. Keeping infections under control is an absolute necessity, and rigorous disinfection protocols serve as our primary line of defense. To effectively neutralize these high-stakes threats, all stakeholders involved in infection prevention and safety should understand pathogen biology and the exact mechanisms of biochemical inactivation.

How does viral structure impact pathogen susceptibility to disinfectants?

Viruses are classified into two primary categories based on their structural composition, which directly dictates their sensitivity to chemical biocides:

  • Enveloped viruses (e.g., Bundibugyo virus (Ebola), Hantaviruses, Influenza, SARS-CoV-2): These pathogens possess an outer lipid membrane. While this layer serves as a structural component, it represents a critical vulnerability. Surface-active disinfectants easily disrupt this lipid bilayer, resulting in rapid inactivation. Though enveloped viruses are typically less virulent (because they don’t always cause cell lysis during cell exit) and yet they have been responsible for some of the most significant headlines and severe epidemics in recent history. Envelope of viruses is derived from host cells originating from Golgi apparatus, ER or Plasma membrane which helps them invade the host immune response and therefore, they have better ability to evade host immune responses and contact spread is more severe. 

  • Non-enveloped viruses (e.g., Polioviruses, Noroviruses, Adenoviruses): These lack a lipid envelope and are protected by a resilient protein capsid. Consequently, they exhibit significantly higher tolerance to standard chemical disinfectants and persist in the environment for extended periods. They are more virulent because they exit the host cell by cell lysis.

How do we choose a disinfectant when viral control is critical?

When evaluating chemical disinfectants across medical, institutional, and industrial sectors, we must analyze the specific risk profile (which includes whether a virus is enveloped vs. non-enveloped). The choice of the right disinfectant should be guided by the type and level of risk involved, as well as by other factors such as disinfectant contact time, material compatibility, and cost. European standards (EN) provide the necessary regulatory framework to ensure products deliver required microbial reduction.

Key EN benchmarks include:

  • EN 14476 (suspension test for virucidal activity): This standard measures a product's capacity to inactivate viruses in a liquid suspension. It offers three tiers of efficacy claims:
  1. Full virucidal activity: covers resilient non-enveloped viruses and all enveloped viruses.
  2. Limited spectrum virucidal activity: covers Adenovirus, Norovirus, and all enveloped viruses.
  3. Virucidal activity against enveloped viruses: covers among others bloodborne and respiratory enveloped viruses such as HIV, HBV, HCV, coronaviruses, Ebola and Hantaviruse.
  • EN 16777 (surface test for virucidal activity): A carrier test evaluating virucidal performance on non-porous surfaces without mechanical action. It bridges laboratory suspension tests and practical application, requiring a minimum reduction (inactivation) of the viral load.
  • Standard for virucidal efficacy determination on hard surface disinfection with mechanical action is under development however will be relying on EN 16615 principles
  • EN 17111 carrier test evaluating virucidal activity for instruments disinfection used in the medical area.

As MVA (Vaccinavirus) remains mandatory test strain for efficacy determination against enveloped viruses in accordance with EN 14476 and EN 16777, products that demonstrated efficacy against this strain in accordance with EN 14476 and/or EN 16777 are considered effective against all other enveloped viruses including Ebola and Hantavirus.
